MacConkey Broth EP/USP, Universal Vial + Durham Tube
A selective and differential liquid medium used to detect, isolate and enumerate coliforms and E. coli in water, milk, and pharmaceutical samples.
Description
0523 – SGL supplies MacConkey Broth EP/USP, Syrup + Durham Tube as a ready-to-use bottled medium for routine microbiological testing. It is suitable for pharmaceutical microbiology, dairy, water testing and clinical testing laboratories. The formulation is described in the European Pharmacopoeia (EP) 2.6.13 and United States Pharmacopeia (USP) <62>.
MacConkey Broth EP/USP is the standard medium for the primary isolation of coliform bacteria. IT is used for the enrichment and detection of Enterobacteriaceae and coliforms. The medium is a selective and differential medium for Escherichia coli spp, Klebsiella spp, Proteus spp, Salmonella, Enterobacteriaceae and coliforms.
Bile salts, which replaced the original sodium taurocholate, inhibit the growth of most Gram-positive organisms. Gelatin peptone supplies nitrogen, vitamins and other nutrients that promote microbial growth.
Lactose provides a fermentable carbohydrate. Lactose-fermenting organisms produce acid, lowering the pH below 6.8 and changing the medium from purple to yellow. Bromocresol purple acts as the pH indicator and turns yellow under acidic conditions. It replaced neutral red because it causes less inhibition. Lactose-non-fermenting organisms, including Salmonella and Shigella species, do not normally acidify the medium or produce this colour change.
An inverted Durham tube captures any gas produced during lactose fermentation. Acid and gas production provide presumptive evidence of coliform growth, potentially including Escherichia coli, but do not confirm the organism’s identity. An acid reaction without detectable gas indicates coliform growth without E.Coli.
Where required, laboratories can subculture the broth onto MacConkey Agar to isolate presumptive colonies before completing appropriate confirmation tests.

A UKAS-accredited laboratory operating to ISO/IEC 17025 (accreditation number 4356) tests the product. At the customer’s request, the laboratory can also perform additional specification testing.
