Description
3134 – Müller Kauffmann Enrichment Broth (MKTTn), Universal is a ready-to-use bottled medium supplied by SGL for routine microbiological testing. It is suitable for use in food and beverage and dairy testing laboratories. Manufactured and quality controlled to SGL internal specifications.
Müller Kauffmann Enrichment Broth (MKTTn)is a selective and differential enrichment broth used for the isolation of Salmonella spp.
The medium originally was said to inhibit lactose fermenting Enterobacteriaceae but not Salmonella. It was modified by Kauffmann to include Ox Bile and Brilliant Green to improve selectivity, and again was later further modified to include novobiocin which inhibits Proteus spp. Gram negative micro-organisms are inhibited.
Due to low numbers of Salmonellae in foodstuff and that many cells are injured during food processing methods, the bacterial cells need to be rescued and pre-enriched in a non-selective broth medium. Once this had been done and incubated at 35-37°C for 18-24hours, a sample can be taken and inoculated into the MKTTn and put back in the incubator for a further 24hours. Presumptive growth needs to be sub-cultured onto XLD and another agar medium for confirmation of Salmonella recovery.
Meat extract and casein provide nitrogen and amino acids; sodium chloride provides osmotic balance. Ox bile, Brilliant Green and Novobiocin are selective agents against non-target micro-organisms. Tetrathionate is formed from the sodium thiosulphate reacting with iodine. Iodine and calcium carbonate buffer the sulphuric acid which is produced from tetrathionate reduction. The calcium carbonate absorbs the toxic metabolites.
SGL manufactures and quality-controls the product to its internal specifications and in compliance with ISO 9001 (reference FM37824). A UKAS-accredited laboratory operating to ISO/IEC 17025 (accreditation number 4356) tests the product. At the customer’s request, the laboratory can also perform additional specification testing.
