Description

0815 – O-F Medium (Hugh & Liefson), Universal is a ready-to-use bottle medium supplied by SGL for routine microbiological testing. It is suitable for use in pharmaceutical microbiology;  cosmetics and personal care. Manufactured and quality controlled to SGL internal specifications.

O-F Medium (Hugh & Liefson) is used to determine if Gram negative bacteria metabolise carbohydrates, such as glucose, oxidatively, by fermentation or have no ability to use the carbohydrates present in the medium (nonsacchrolytic).

Two tubes are tested per organism, with one of the bottles being overlayed with mineral oil. This overlay prevents diffusion of oxygen into the medium, therefore creating an anaerobic environment within the tube.

The medium has a high concentration on carbohydrates, and a lower concentration of peptone to avoid the chances of an aerobic organism utilising peptone, which would neutralised the limited acid production from the oxidative organism.

Bacteria that can ferment carbohydrates produce an acidic reaction in both the aerobic and anaerobic tube. This acid production caused the medium to change from a blue-green colour to yellow. Non-fermenting bacteria that metabolise carbohydrates by oxidation produce a small amount of acid in the aerobic tube with no reaction in the anaerobic tube.

Dipotassium phosphate is used as a pH controlling buffer as well as it’s ability to promote fermentation. Sodium chloride provides essential electrolytes for transportation of the organisms and offers osmotic balance. The agar aids in distribution of acid throughout the medium.

SGL manufactures and quality-controls the product to its internal specifications and in compliance with ISO 9001 (reference FM37824). A UKAS-accredited laboratory operating to ISO/IEC 17025 (accreditation number 4356) tests the product. At the customer’s request, the laboratory can also perform additional specification testing.
