0591 – Selenite F Broth — Double Strength, allows larger amount of samples to by analysed.

Selenite F Broth  – Double Strength selectively enriches Salmonella from foodstuffs, water, faeces and other sample types. The double-strength formulation enables laboratories to analyse larger sample volumes.

Selenite F Broth combines buffered lactose broth with sodium biselenite. The “F” refers to faeces, as laboratories commonly use the medium to isolate Salmonella and Shigella from faecal specimens. Sodium biselenite inhibits many Gram-positive and Gram-negative bacteria, including coliforms and enterococci, by reacting with sulfhydryl groups in microbial cells. Users must handle sodium biselenite with extreme care because of its toxicity and potential teratogenic effects (due to it reacting with sulfhydryl groups in microbial cells). Sodium phosphate stabilises the pH of the medium and reduces the toxicity of the selenite.

Laboratories should not use Selenite F Broth as a standalone medium. After the initial 18–24-hour incubation, they should subculture the broth onto a Salmonella-selective agar, such as Xylose Lysine Deoxycholate (XLD) Agar or Hektoen Enteric Agar. Presumptive Salmonella typically produces pink or red colonies with black centres on XLD Agar and blue-green colonies, often with black centres, on Hektoen Enteric Agar.
